چکیده

Background/objective — The goal of our study was to investigate the role interaction between polymorphic loci immune response mediator genes (TNFA rs1800629, LTA rs909253, IL1B rs16944, IL2-IL21 rs6822844, IL2RA rs2104286, IL6 rs1800795, IL10 rs1800872, MIF rs755622, CTLA4 rs3087243, NFKB1 rs28362491, PTPN22 rs2476601, and PADI4 rs2240336) in formation a genetic predisposition juvenile idiopathic arthritis (JIA). Material Methods involved 330 JIA patients 342 volunteers from Republic Bashkortostan. Genotyping conducted via real-time polymerase chain reaction. gene-gene interactions were studied using multifactor dimensionality reduction algorithm. Results In general analysis, best model combination rs16944 – rs1800872 rs28362491 rs2240336 loci. However, after gender-based stratification results obtained when examining combinations rs1800795 girls rs2104286 boys. Within all these models, genotype associated with both augmented reduced risks identified (taking into account gender-specific differences). Conclusion implied that an important is played by

Immune Complexes in Juvenile Idiopathic Arthritis

Juvenile idiopathic arthritis (JIA) reflects a group of clinically heterogeneous, autoimmune disorders in children characterized by chronic arthritis and hallmarked by elevated levels of circulating immune complexes (CICs) and associated complement activation by-products in their sera. Immune complexes (ICs) have been detected in patients' sera with JIA utilizing a variety of methods, including...

Multicentric Carpotarsal Osteolysis Mimicking Juvenile Idiopathic Arthritis

Background Multicentric carpotarsal osteolysis (MCTO), a skeletal dysplasia presents in early childhood mimicking juvenile idiopathic arthritis (JIA). Recognition of this syndrome is essential to avoid unnecessary treatment with immunosuppressive agents because of different course and treatment. Case Report A 3-year-old boy presented with swelling and restriction of right wrist joint and left ...

Predictors of response to methotrexate in juvenile idiopathic arthritis

BACKGROUND The response to methotrexate so far is unpredictable in patients with juvenile idiopathic arthritis. Thus such predictors have to be determined in a large patient cohort. METHODS Demographic, clinical, articular and laboratory variables of patients newly treated with methotrexate were analysed by bivariate and logistic regression analysis to identify predictors of response to metho...

Autoimmune response to transthyretin in juvenile idiopathic arthritis.

Juvenile idiopathic arthritis (JIA) is the most common pediatric rheumatological condition. Although it has been proposed that JIA has an autoimmune component, the autoantigens are still unknown. Using biochemical and proteomic approaches, we identified the molecular chaperone transthyretin (TTR) as an antigenic target for B and T cell immune responses. TTR was eluted from IgG complexes and aff...

Autoinflammatory genes and susceptibility to psoriatic juvenile idiopathic arthritis

OBJECTIVE To investigate the association of NLRP3, NOD2, MEFV, and PSTPIP1, genes that cause 4 of the autoinflammatory hereditary periodic fever syndromes (HPFS), with juvenile idiopathic arthritis (JIA). METHODS Fifty-one single-nucleotide polymorphisms (SNPs) across the 4 loci were investigated using MassArray genotyping in 950 Caucasian patients with JIA living in the UK and 728 ethnically...
